A new flow cytometric assay for the evaluation of phagocytosis and the oxidative burst in whole blood

Abstract

Phagocytes play an important role in host defence against microorganisms and different techniques are needed to evaluate their functional activities. Here we describe a rapid, simple and reliable one step procedure to measure the phagocytosis rate and oxidative burst activation of both polymorphonuclear leukocytes (PMN) and monocytes, by means of flow cytometry using only small quantities of whole blood. The two species of bacteria employed as test microorganisms, Staphylococcus aureus and Candida albicans showed a somewhat different behaviour. We found that the present method could be used as an alternative test in the diagnosis of chronic granulomatous disease (CGD). Moreover we were able to analyse, in a one step procedure, defective phagocyte functions in a group of paediatric patients suffering from recurrent microbial infections.

    We observed an increased production of ROS in D384 cells after TiO2NPs using DHE, a cell permeable fluorescent dye that is oxidized by ROS present in the cytoplasm to ethidium bromide, a nucleic acid stain. The red fluorescence intensity in the nucleus corresponds to the amount of ROS inside the cells (Bucana et al., 1986; Perticarari et al., 1994; Shimoni et al., 2006). Thus, elevated ROS following nano-TiO2 treatment, as observed in these human derived astrocytes, may induce apoptosis in brain cells since oxidative damage to DNA and membranes has shown to induce the apoptosis.

    The blood from the fish was then obtained from the caudal vein and the fish was dissected for further analysis. Three replicate fish from each treatment were quickly dissected for the spleen to measure the phagocytic index, which was carried out using a protocol modified from Perticarari et al. (1994).     After a wash with cold HBSS, the bacteria were resuspended in HBSS (1 × 108 cells mL−1) and stored in the dark at 4 °C. Based on the protocol described by Perticarari et al.
