Protocols utilizing proteases such as alpha-amylase, collagenase, and trypsin combined with reducing and chaotropic agents have been described (Daamen et al., 2001; John & Thomas, 1972; Rasmussen, Bruenger, & Sandberg, 1975; Starcher & Galione, 1976; Steven & Jackson, 1968). Below are two methods that utilize trypsin and cyanogen bromide digestion (because elastin lacks methionine, cyanogen bromide is used to cleave nonelastin proteins).
