Purification and comparison of elastins from different animal species

Abstract

A nondegradation method was developed for the isolation of elastin from tissues of 10 different animal species. Marked differences, involving primarily the neutral and aromatic amino acids, occurred in the amino acid composition of elastin of different species. The amino acid composition of elastins from a given species was constant for all the tissues studied.

    Therefore, several approaches have been developed to minimize elastin damage by incorporating enzyme treatment and reducing agents into the extraction protocol. Protocols utilizing proteases such as alpha-amylase, collagenase, and trypsin combined with reducing and chaotropic agents have been described (Daamen et al., 2001; John & Thomas, 1972; Rasmussen, Bruenger, & Sandberg, 1975; Starcher & Galione, 1976; Steven & Jackson, 1968). Below are two methods that utilize trypsin and cyanogen bromide digestion (because elastin lacks methionine, cyanogen bromide is used to cleave nonelastin proteins).

    The results were expressed as percentage of tissue dry weight. It should be noted that the amount of desmosine and isodesmosine in mature elastin varies in different tissues and species [42] and that the adopted conversion value above was derived from human aorta and lung. Total content of sulfated glycosaminoglycans (GAG) was measured using the 1,9-Dimethylmethylene Blue assay as previously described [43].
