The study included 112 patients with mild-to-severe stable COPD patients and 32 healthy volunteers. Cytokine levels (IL4, IL6, IL17A, IFNγ, TGFβ1, FGF, TNFα) and granulocyte percentage in the blood were determined by flow cytometry (BD FACSCantoII). Spearman's coefficient (r) was assessed, statistical significance was p<0.05. In mild COPD, an increase in CD282+, CD284+ granulocyte percentage by 18% and 55% compared with the control was found. An elevated CD282+ cell number was associated with increased TNFα (0.85), IL17A (0.80), IFNγ (0.74) levels. The number of CD282+, CD284+ granulocytes was 40% and 91% higher in moderate COPD, than in the control group. Correlations between CD282+ cell number and the content of TGFβ1 (0.91), TNFα (0.77), IL17A (0.73), IFNγ (0.63), IL6 (0.54); between CD284+ cell percentage and IL4 (0.59), TGFβ1 (0.55), FGF (-0.54), IL10 (0.63), IL17A (0.45) levels were revealed. In severe COPD, CD282+/CD284+ cell number exceeded the control values by 71% and 124%. CD282+ cell number correlated with TGFβ1 (0.96), IL6 (0.94), IL17A (0.93) levels; CD284+ cell number correlated with the content of IL4 (0.8), TGFβ1 (0.66), FGF (-0.95), IL10 (0.8), IL17A (0.68), IL-6 (0.57). Thus, as COPD progresses, an increase in the number of CD282+/CD284+ granulocytes is associated with an elevation of levels of anti-inflammatory cytokines (IL4, IL10), Th17 cytokines (IL6, IL17A, TGFβ1), a decrease in FGF level involved in airway remodeling.FootnotesCite this article as: European Respiratory Journal 2021; 58: Suppl. 65, PA3423.This abstract was presented at the 2021 ERS International Congress, in session “Prediction of exacerbations in patients with COPD”.This is an ERS International Congress abstract.
