But, the mechanism of tigecycline resistance were not very clear.Methods: In vitro evolution of tigecycline-resistant Klebsiella pneumoniae (KP), the minimum inhibitory concentrations (MICs) of tigecycline was determined by micro-broth dilution method. RNA sequencing and data analysis were performed for investigating differentially expressed genes. RT-PCR were perform for verifying the differentially expressed genes.Results: Three groups toll 9 strains of KP were involve in our study, Strains A2 and A3 were induced from A1, B2 and B3 were induced from B1, C2 and C3 were induced from C1. The tigecycline MICs of A1,B1 and C1 were 0.5 μg/mL; A2, B2 and C3 were 16.0 μg/mL; A3, B3 and C3 were 32.0 μg/mL. RNA-sequencing and RT-PCR confirmed that cusE, cusS, cusR, cusC, cusF, cusB and cusA were higher expression among C2 or C3 than C1.Conclusion: CusS-CusR two component system was not only associated with bacteria resistant to copper/silver, but also associate with antibiotic resistance, we speculated that cusS-cusR two component system was associated with CRKP resistant to tigecycline.FootnotesCite this article as: European Respiratory Journal 2019; 54: Suppl. 63, PA4565.This is an ERS International Congress abstract.
