RT Journal Article SR Electronic T1 The associations between a smoking and IL-10 gene polymorphism in patients with MDRTB JF European Respiratory Journal JO Eur Respir J FD European Respiratory Society SP PA2730 DO 10.1183/13993003.congress-2016.PA2730 VO 48 IS suppl 60 A1 Dmytro Butov A1 Mykhailo Kuzhko A1 Tetyana Butova YR 2016 UL http://erj.ersjournals.com/content/48/suppl_60/PA2730.abstract AB Background and objective: Determine the associations between a smoking and IL-10 gene polymorphism in patients with MDRTB in lungs on the background of the respective IL-10 production of blood.Methods: The study included 170 Caucasian people with infiltrative pulmonary TB permanently residing in Ukraine including 60 patients with MDRTB and smoking(1stgroup),36-without MDRTB and smoking(2ndgroup),14-with MDRTB and non-smoking(3rdgroup )30-without MDRTB and non-smoking(4thgroup)and 30 healthy donors (5thgroup).Studied promoter region G1082A of IL-10 gene(rs1800896).Serum levels of IL-10 were evaluated by ELISA.Results: In the 1stgroup the levels of IL-10 were(31.11±1.16)pg/L,2nd-(35.39±1.01)pg/L,3rd-(38.27±0.88)pg/L,4th-(42.39±0.92)and5th-(50.25±1.26)pg/L(p<0.05). Homozygote GG genotype predominated in 5thgroup was 56.67±9.05%(N=17) compared to patients with TB: 1st group-3.33±2.32%(N=2), 2nd-2.78±2.74%(N=1), 3rd-7.14±7.14%(N=1) and 4th-26.67±8.07%(N=8).Heterozygous GA genotype was observed in95.00±2.81%(N=57)in1st,25.00±7.22%(N=9) in 2nd,4.29±9.71%(N=2) in the 3rd,6.67±4.55%(N=2) in 4thand 23.33±7.72%(N=7) in 5thgroups.Homozygous AA genotype was 1.67±1.65%(N=1) in the 1stgroup,in 2nd-72.22±7.47%(N=26), 3rd-78.57±11.38%(N=11),4th-66.67±8.61%(N=20)and5th-20.00±7.30%(N=6)(p<0.05).Conclusion: Smoking contributes to significant activation polymorphism G1082A gene IL-10 heterozygous GA genotype that may lead to changes in the immune system making susceptible to MDRTB.Compared to healthy controls patients with TB had significantly decreases levels of serum IL-10.This coincided with greater frequency of heterozygous GA and homozygous AA genotype polymorphism G1082A of the IL-10 gene.
