However, its role in lower airway immunity has not been yet satisfactory explained.Aims: Assessment of IL27 expression in lower airway pathology, including interstitial lung diseases (ILD) and non-small cell lung cancer (NSCLC).Methods: IL-27 expression was examined by ELISA in BAL supernatants from patients with pulmonary sarcoidosis, idiopathic pulmonary fibrosis, nonspecific interstitial pneumonia, EAA, NSCLC and controls (n=22, 9, 8, 9, 13 and 14, respectively). In oncological patients lavage was harvested from the suspicious lung region. BAL cells were immunostained for intracellular IL27 expression, AL subsets were phenotyped. Real-time PCR was performed to prove IL27 expression in AL.Results: IL27 level was significantly increased in EAA nonsmokers (6.8±1.6 vs 1.6±0.8ng/ml in controls, p<0.01) and in NSCLC smokers (16.4±13.4 vs 4.0±0.9ng/ml in controls, p<0,05). Parallel results were shown for NSCLC nonsmokers (40,0±0.9ng/ml). However the elevation of IL-27 level was insignificant (low number of tested subjects). IL-27 level elevation in PS was insignificant p<0,1). No changes were observed in other ILD. We found AL IL-27+ in all tested groups and proved IL-27 transcription in AL. IL-27 expression was not correlated with any AL subset, including Th, Tc, either nTreg cells.Conclusions: IL27 is produced in lower airways by lung lymphocytes. IL-27 highly increased expression in NSCLC suggests its role in anti-tumor immune response. IL-27 should be tested as experimental tool in patients with lung cancer. ER -
