The objective of this study is to identify new genes which is useful for evaluating pulmonary inflammation induced by NiO. Rats were exposed to 0.2mg or 1mg of NiO and total RNAs were isolated from lung at 3 days, 7 days, 1 month, 3 months, 6 months, 12 months and 24 months after the end of exposure. Analysis of cDNA microarray was performed with total RNA at 1 month after the end of exposure and revealed that 450 kinds of mRNAs were up-regulated over 2 times and 388 kinds of mRNAs were down-regulated compared with control. Among up-regulated 450 mRNAs, 16 kinds of mRNAs were up-regulated over 8 times. Six of 16 kinds of mRNAs (Ccl2,Ccl7,Cxcl-5,Cxcl-10,Cxcl-11 and Mmp12) were involved in inflammation and investigated until 24 months post-exposure by real-time PCR. Expression peak is different in each gene, but gene expression levels of Ccl2,Ccl7,Cxcl-10 and Cxcl-11 were decreased to almost same level of control and those of Cxcl-5 and Mmp12 were continuously up-regulated until 24 months. These 6 genes might be useful markers for evaluation of pulmonary inflammation by nickel oxide nanoparticles.
