After description of smoking history and signed consent, 10 puffs (3 sec.) were scored, one every 5 minutes with the 10 different combinations.Results: Responses from 350 puffs from smokers aged 22.0±7.7years, shown:• The optimal throat-hit was obtain after a learning process of 5 puffs.•The Fagerström miniscore is link to nicotine concentration giving optimal throat-hit with a1.6 mg/ml nicotine increase for each Fargerström score point.•The score of throat-hit is correlate to the desire to quit smoking to e-cigarette (R2 = 0.94). This desire is<1/10 when throat hit is unpleasant (score 0-3) and exceeds 7/10 when throat hit is pleasant (score 7-10).• The nicotine concentration associated with optimal throat hit is 7 times 0 mg/ml and 9 times 18mg/ml with 3.5 volts. For the 35 smokers, a nicotine concentration of 12.6 mg/ml with 3.5 volts and 10.89 mg/ml with 4.5 volts is associated with optimal throat hit.Conclusions: This pilot study show a link between Fargerström miniscore and the concentration of nicotine necessary to obtain an optimal throat-hit with e-cigarette. The strong relationship between the optimal throat-hit and the desire to use e-cigarette to quit smoking, justifying to improve e-cigarettes technology and e-liquid to help smokers to finds optimal throat-hit.
