Abstract
Goal: to evaluate the protective effect of inhaled glucocorticosteroid (ICS) beclomethasone dipropionate (BDP) and long-acting beta2-agonist (LABA) formoterol (F) combination on COPD model.
Materials and methods: COPD model was reproduced in rats during a discrete 60-day exposure of nitrogen dioxide. Starting from day 61, for 30 days the animals of the experimental group (n = 13) were daily inhaled through a special mask with an aerosol combination of ICS BDP and LABA F (Chiesi Farmaceutici, Italy). Control group (n = 13) received placebo. Cell composition and cytokine-enzyme profile of the BALF, content of secretory immunoglobulin A (sIgA) and surfactant protein D (SP-D) were determined by ELISA. Histological examination of lung tissue was performed.
Results: Under the influence of BDP+F inhalations, activity of neutrophilic lymphocytic inflammation decreased (H - 7.4 ± 1.2% and L - 10.3 ± 1.8%, p <0.05), the profile of cytokines and enzymes in BALF normalized: TNFa (23.1 ± 1.1 vs 32.5 ± 3.1 pg/ml in the control, p <0.05), IL-8 (25.5 ± 1.2 vs 41.2 ± 2.6 pg/ml, p <0.05), IL-17 (43.8 ± 6.1 vs 98.7 ± 5.4 pg/ml, p <0.05), NE (18.7 ± 1.2 vs 33.2 ± 2.5 ng/ml, p <0.05), MMP-12 (1.7 ± 0.1 vs 2,7 ± 0.1 ng/ml, p <0.05). Morphological structure of the bronchoalveolar epithelium was restored and, as a result, its functional activity indicators improved: sIgA (38.6 ± 3.7 vs 11.5 ± 1.3 μg/mg protein in the control, p <0.05) and SP-D (94.8 ± 6.5 vs 44.2 ± 5.2 pg/ml, p <0.05).
Conclusion: The use of ICS BDP and LABA F combination on COPD model had a pronounced anti-inflammatory effect, coupled with the activation of physiological repair processes in lungs.
