Abstract
Carcinoma-associated fibroblast (CAF) as prominent cell type of the tumour microenvironment has complex interaction with the cancer cells. Our group has proved that Farnesoid X receptor(FXR ) promoted tumor growth via directly transactivating CCND1. This study was aimed to explore the possible impact of FXR expression on the proliferation and migration of CFA. Four strains of CAF (two separately from lung adenocarcinoma and squamous cell carcinoma) were isolated by means of differential attachment technique and forwarded for morphologic examination. Human pulmonary fibroblasts (HPF) was choosed as normal pulmonary fibroblast. The resultss showed that proliferation of the four CAF was significantly higer than HPF(P<0.01)(Fig1-a) by cell proliferation assay. When cocultured with A549 mock or A549 cell with high expression of FXR( FXR hi), the proliferaqtion of most significant in A549 FXR hi group, followed by A549 mock group, the difference among these three groups were significant(P<0.01). Compared with HPF,the cell counts that transed membrane of CAF groups was much more than HPF group. Western blot revealed that the expression of α-SMA、FAP 、FXR in CAF was elevated . Cell cycle assay showed the ratio of S stage in CAFs was higher than HPF (P<0.01). All of these suggest an activation of CAF. It is concluded that A549 cell can promote the proliferation and migration of fibroblast through FXR signal pathway .(sponsored by National natural Science Foundation of China 81874314 )
