Abstract
The aim of this study was to investigate the association between polymorphisms in IL17A and IL17F and asthma control. This work was performed with participants from ProAR case-control study in Salvador, Brazil. Asthma control was defined according to Global Initiative for Asthma. In this work were included 209 subjects with controlled asthma, 522 with poor control of asthma and 225 with uncontrolled asthma. The rs2275913, rs3819024, rs763780 and rs2397084 were genotyped in genomic DNA of these individuals using TaqMan probe- based 5´-nuclease assays technology. Logistic regressions were performed using PLINK software 1.9, using the three genetic models, adjusted for sex, age, and self-reported skin color. Linkage disequilibrium and haplotypes analysis were made using Haploview 4.0 and SNPstats. In order to evaluate the functional impact of such variants, IL-17A was measured in sputum as well as cells expressing IL-17 was explored by flow cytometry. In silico analysis were also conducted to further investigate functional variants using rSNPbase, SNPeffect3D, I-Mutant, Hope, SHIFT and Polyphen 2.0. The allele A of rs2275913 was a risk factor for uncontrolled asthma (OR:1.79;95%;CI:1.22-2.73–additive model) and the G allele of rs3819024 (OR:2.19;95%CI:1.04-4.06–recessive model) and the A allele of rs2275913 (OR:1.33;95%CI:1.31-1.99–recessive model), were risk for poor controlled asthma, respectively. In silico analysis have indicate that such SNVs may be involved in several mechanisms of gene expression regulation. No association was found between variants and IL-17A measured in sputum as well as cells expressing IL-17. Genetics variants in IL17A and IL17F may influence asthma control in our study population.
