Abstract

Leukotrienes (LTs) play a central role in asthma. Moderate intensity aerobic exercise reduces asthmatic inflammation. Then, this study investigated if the anti-inflammatory effects of aerobic exercise is due to inhibition of LTs pathway.Forty male BALB/c mice were distributed in Control, Exercise, OVA and OVA+Exercise groups. Mice were sensitizedand challenged using ovalbumin. Moderate intensity treadmill training was performed 5x/week, 1h/session, for 4 weeks.Aerobic exercise decreased the number of eosinophils (p<0.001), neutrophils (p>0.001), lymphocytes (p<0.001) and macrophages (p<0.01) in BAL, as well as the number of eosinophils (p<0.01), lymphocytes (p<0.001) and macrophages (p>0.001) in airway wall. Also, airways collagen (p<0.01) and elastic fibers (p<0.01) content, mucus production (p<0.01) and smooth muscle thickness (p<0.01) was reduced in exercised mice. In addition, BAL levels of IL-4 (p<0.01), IL-5 (p<0.01) andIL-13 (p<0.01) was reduced, as well as the levels of CysLT (p<0.05), LTB4 (p<0.01), while IL-10 was increased in Exercised and OVA+Exercised groups (p<0.01). The expression of 5-LO (p<0.05), LTA4H (p<0.05), CysLT1 receptor (p<0.001), CysLT2 receptor (p<0.001), LTC4 synthase (p<0.001) and BLT2 (p<0.01) by peribronchial leukocytesand by airway epithelium was reduced by exercise, and also the expression of BLT2 and CysLT2 receptor in lungs proteins extract. Finally, AHR was also reduced in OVA-trained mice at MCh 25mg/mL (p<0.05) and MCh 50mg/mL (p<0.05). Moderate intensity treadmill training reduces asthma phenotype by inhibition of leukotrienes pathway.
